Pdx1 is essential for pancreatic progenitor
formation/differentiation. However, it remains unknown how
the very dynamic patterns of Pdx1 expression, regulated via
specific cis-regulatory modules, are linked to is function.
The compact, highly conserved 5’ Area II region contains binding
sites for multiple endocrine/beta cell transcription factors and
has beta-cell-specific promoter activity. We hypothesized
that Area II is the primary control element for Pdx1 in
differentiating and mature beta cells. We tested this idea
genetically by generating mouse mutants carrying Area II
deletion. I will discuss how removal of Area II results in
large transcriptional and epigenetic influences on the endocrine
program, with strong effects in endocrine specification and beta
cell maturation.
