Meeting Agenda/Notes

Unlike small molecule or biologic drugs, cells can integrate multiple inputs to perform an array of outputs as diverse as the assembly of neotissue or engulfment of foreign bodies. As such, cells can serve as exceptional agents to coordinate regeneration and combat disease. Wedding synthetic biology, the field that attempts to rationally assemble new biological systems that possess predictable and finely controlled properties, with cell therapies has resulted in remarkable outcomes in certain medical specialties, such as cancer immunotherapy. In this seminar, I will present on our efforts to realize a vision of treating complex diseases, particularly of the musculoskeletal and central nervous systems, with such living therapeutic devices. I will also discuss nascent areas of our research program, which focus on developing high fidelity organoid models of human systems by genetically programming stem cells to reliably self-assemble and faithfully develop into target tissue mimetics. By combining principles from synthetic biology, genome engineering, and regenerative engineering, our approach potentiates the generation of tissue replacements with biomimetic features of native tissues while precisely defining how cells respond to cues they encounter in development and in pathological microenvironments.
