Determining the genetic and epigenetic factors involved in differentiation and maintenance of pancreatic cell lineages will be instrumental for developing targeted therapies. In our prior work, we combined cell purification methods with genomic assays to identify cell-type specific accessible genomic regions in primary human pancreas cells, and found putative enhancer elements corresponding to specific pancreas cell lineages. However, the mechanisms linking these regions to cell type-specific gene expression remained unclear. In my talk, I will present our new work on Hi-C linked with chromatin immunoprecipitation (HiChIP) assays using purified human pancreas cells to elucidate chromatin loops involving enhancer elements, as well as our efforts to perturb enhancer function in beta cells.
