We have discovered a dosage-dependent and cooperative role of Pdx1 and Oc1 in specification of the endocrine lineage. Heterozygosity for both Pdx1 and Oc1 results in a reduction in the number of hormone-positive cells during development and subsequent impairment in beta-cell function postnatally.
