I will describe a collaborative effort that resulted in the generation of a comprehensive lineage-directed gene correlation network for mouse β-cells. This undertaking utilized a total of approximately 1.5 billion mapped sequence reads from 33 RNA-Seq datasets representing eleven different FACS-purified murine cell populations, each of which were temporally and genetically defined. Our study required the development of iterative WGCNA approach which, in the end, enabled us to classify 10,721 of the 15,949 genes expressed in these cell populations into 79 modules. We further grouped these modules using community detection analysis into 14 meta-modules. Together, this effort resulted in an easily interpretable framework for the development of pancreatic β-cells and related functional predictions.
