The focus of my laboratory is to understand the cellular and molecular regulation of the alveolar capillary barrier during acute infection and injury. I aim to uncover novel mechanisms that contribute to the development of barrier dysfunction in sepsis and ARDS with the ultimate goal of identifying new biologic targets and unique therapeutic approaches that can be translated to clinical practice. My research approach is translational, utilizing patient samples and clinical data, mouse models and cell lines to study fundamental biologic mechanisms that regulate lung responses to injury. My molecular focus is on cell-free hemoglobin and Tissue Factor and my disease focuses are ARDS and sepsis. I also have a special interest in the biologic underpinnings of gender differences in acute illness, sepsis and ARDS. My goal is to translate findings from these mechanistic studies to develop personalized care for the critically ill patients. Much of my work has focused on lung permeability as it is regulated by the epithelium and endothelium. I have extensive experience with mouse and cell culture models of lung injury and currently have over ten murine lung injury models established in my laboratory. My laboratory is currently funded with two R01’s studying the effects of cell-free hemoglobin in sepsis and ARDS, a VA Merit Review defining the non-coagulant and protective role of alveolar epithelial Tissue Factor and a Department of Defense grant to study the lung epithelial glycocalyx in exhaled breath from ARDS patients. In addition, I have several ongoing projects using the Synthetic Derivative and BioVU to understand clinical and genetic underpinnings of acute and chronic lung diseases. I am currently a mentor on Gordon Bernard, Timothy Blackwell’s and Eric Delpire's T32 training grants. I have mentored 21 students, residents and fellows and currently serve as mentor for both a clinical fellow on this T32 and a junior faculty member who currently holds three mentored grants including the prestigious Parker B. Francis Fellowship in Pulmonary Medicine. My lab is always open to interested trainees and scientists at all levels interested in exploring important biological and clinical questions using a multitude of basic and translational research tools.
