Our laboratory focuses on the development of intestinal immune regulation in preterm infants. Premature infants are at risk for severe inflammatory complications such as necrotizing enterocolitis (NEC). I am a board-certified Pediatrician with additional post-doctoral training in Pediatric Infectious Diseases and Neonatal-Perinatal Medicine. For my post-doctoral training I created new technologies to compare the genetic antibody repertoire in virus-specific B cells between infants and adults. I expanded my research to study the development of T regulatory cells in the intestine of premature infants. I developed the collaborative infrastructure for the efficient procurement of fresh intestinal surgical tissue specimens from preterm infants with gastrointestinal pathology and by establishing methods for the analysis of single viable lymphocytes from these tissue sections. This surgical tissue repository has been a critical resource for human NEC tissue for numerous collaborators in the nation.  Our laboratory developed tools that for the first time permit the study of the ontogeny and function of immune effector and suppressor cells in the premature intestine. To overcome the ethical and technical limitations of human immunology research in this population, we customized multicolor flow cytometry protocols and microfluidic cell trap devices to study single immune cells isolated from surgically resected tissue specimens from infants with and without NEC. Using simultaneous sampling of intestinal tissue and feces during surgical resection, we have now established how the intestinal microbiome differs between the mucosal surface and stool, reflective of the distinct biogeography of the neonatal gastrointestinal tract and determined the specific intestinal microbiome profile in preterm infants with NEC
